He developed some among the first NGS analysis pipelines and quality control tools. Riccardo started his career in the Sardinian Genome Study stemming from the ProgeNIA project, on which he worked for his PhD thesis. His results were published with the collaboration on the journal Science.  In 2013 he moved to the Helmholtz Center in Munich for his postdoc with Prof. Thomas Meitinger and Dr. Tim Strom and continued at the Institute of Human Genetics (IHG) of the University Hospital of the Technical University of Munich (TUM), where he was head of the Bioinformatics group till December 2023 and so far produced around 60 peer reviewed publications. At the IHG he managed NGS data analysis applications for research and diagnostics. In the clinical field he acquired a strong experience as software developer, in process control, and in quality management, contributing to the ISO15189 certification of the Institute as a diagnostic laboratory, covering laboratory processes and the bioinformatic analyses developed in house. He is responsible of the development of the open source software EVAdb, for the clinical interpretation of the variants from human genomes and exomes and RNAseq. He also manage the development of their associated pipelines and of the EVAdb LIMS. The goals of the project include studying the effect of RT on melanoma and healthy tissues such as skin, adipose tissue, heart, and lung in mice. This will be accomplished by injecting melanoma cells into mice and then exposing them to RT. Current research on FLASH therapy suggests that although there have been some studies on the effects of this treatment on skin tissue, there is still a significant knowledge gap regarding its impact on heart and white adipose tissue (WAT). This lack of information leaves important questions about the safety and efficacy of FLASH therapy unanswered. Considering the vital functions that the heart and WAT perform in the body, it is critical to fill this gap in our knowledge to ensure that FLASH therapy is a safe and efficient treatment option for patients. Therefore, further research is urgently needed to provide a comprehensive analysis of the impact of FLASH therapy on these tissues.\nHistopathological, transmission electron microscopy, and molecular analyses to study markers of damage and function will be used to create a database to instruct an artificial intelligence (AI) to identify patterns of damage and function in healthy and tumor tissues, with the goal of improving the efficacy of RT and reducing its side effects.\nExpected results include a better understanding of the effect of RT on melanoma and healthy tissues, which could lead to new therapeutic strategies for the treatment of melanoma. The project is divided into 3 Work Packages: - WP1 activities will be aimed at the development of laboratory protocols and bioinformatics pipelines to be able to respond to all the needs of genetic-molecular characterization of the transcriptome and genome; - WP2 involves the validation of the workflow including all protocols and pipelines developed by performing replicate experiments on samples with known results; - WP3 will consist of the application of the workflow on samples that will be sent to the CRS4 NGS platform with a flow of 6 patients per month.\n\u00a0\nProtocols based on NGS technology including structural changes (short read technology) and post-transcriptional chemical modifications (native RNA\/DNA long read technology) of coding and non-coding RNAs will be developed for comprehensive expression profile analysis.\n\u00a0\nGenome sequencing will provide useful information to elucidate the origins, genetic and epigenetic, of neoplastic transformation of cells.
